چکیده

This paper reports an electron microscope examination of the cortices of some plant cells engaged in wall formation. Previous studies of similar material fixed in OSO(4) alone have disclosed discontinuities in the plasma membrane and other evidence of inadequate fixation. After glutaraldehyde, used as a fixative in this present study, the general preservation of cortical fine structure is greatly improved. This is shown, for example, by the first evidence of slender tubules, 230 to 270 A in diameter and of indeterminate length, in plant cells of this type. They have been found in the cortical regions of cells of two angiosperms and one gymnosperm, representing all the material so far studied following this method of fixation. The tubules are identical in morphology to those also observed here in the mitotic spindles of plant cells, except that the latter have a somewhat smaller diameter. It is noted that the cortical tubules are in a favored position to govern cytoplasmic streaming and to exert an influence over the disposition of cell wall materials. In this regard it may be of some significance that the tubules just beneath the surface of the protoplast mirror the orientation of the cellulose microfibrils of the adjacent cell walls.

Pap1+ confers microtubule damage resistance to mut2a, an extragenic suppressor of the rad26:4A allele in S. pombe.

The DNA structure checkpoint protein Rad26ATRIP is also required for an interphase microtubule damage response. This checkpoint delays spindle pole body separation and entry into mitosis following treatment of cells with microtubule poisons. This checkpoint requires cytoplasmic Rad26ATRIP, which is compromised by the rad26:4A allele that inhibits cytoplasmic accum...

Microtubule Fine Structure

Recent studies have demonstrated the widespread occurrence of microtubules in the cytoplasm of both animal and plant cells (e.g., Burgos and Fawcett, 1955; Manton , 1959; Slautterback, 1963; Ledbetter and Porter, 1963). The microtubules have usually been examined in sectioned material after osmium tetroxide or glutaraldehyde fixation. Because of advantages in the negative staining technique, it...

Fine-Tuning Plant Growth in the Face of Drought.

Limiting shoot growth is an important survival strategy for plants during times of drought; smaller leaves mean that less water is lost through transpiration and more is retained in the soil. As drought stress restricts both cell division and expansion – processes that rely on specialized microtubule arrays – factors that regulate microtubule organization and stability likely affect a plant’s a...

Effects of T208E activating mutation on MARK2 protein structure and dynamics: Modeling and simulation

Microtubule Affinity-Regulating Kinase 2 (MARK2) protein has a substantial role in regulation of vital cellular processes like induction of polarity, regulation of cell junctions, cytoskeleton structure and cell differentiation. The abnormal function of this protein has been associated with a number of pathological conditions like Alzheimer disease, autism, several carcinomas and development of...

Immunocytochemical Study on Microtubule Reorganization in HL-60 Leukemia Cells Undergoing Apoptosis

Background: Microtubules (MT) are important components of cell cytoskeleton and play key roles in cell motility mitosis and meiosis. They are also the targets of several anticancer agents which indicating their importance in maintaining cell viability. Microtubular reorganization contributing to apoptotic morphology occurs in normal and neoplastic cells undergoing apoptosis induced by cytotoxic...

In-silico Investigation of Tubulin Binding Modes of a Series of Novel Antiproliferative Spiroisoxazoline Compounds Using Docking Studies

Interference with microtubule polymerization results in cell cycle arrest leading to cell death. Colchicine is a well-known microtubule polymerization inhibitor which does so by binding to a specific site on tubulin. A set of 3',4'-bis (substituted phenyl)-4'H-spiro[indene-2,5'-isoxazol]-1(3H)-one derivatives with known antiproliferative activities were evaluated for their tubulin binding modes...
